Anti-hyperuricemia and renal protection effects of total flavonoids from corn silk
Aim:To explore the value of total flavonoids from corn silk(TFCS)in the treatment of hyperuricemia(HUA)and the protection of kidneys in rats.Methods:A total of sixty SD rats were randomly allocated into normal control group,model group,allopurinol group,and TFCS low,medium,and high-dose groups,with 10 rats in each group.The normal control group was given physiological saline by gavage,while the other groups were treated with 250 mg/(kg·d)ethambu-tol and 100 mg/(kg·d)adenine suspension daily to construct the rat HUA model.After molding,the allopurinol group was given 50 mg/(kg·d)allopurinol,while the low,medium,and high-dose TFCS groups were given 0.5,1.0,and 2.0 mg/(kg·d)TFCS by gavage for 14 days.The general state of rats was observed and recorded,and the weight of rats in each group was recorded.After the last administration,serum uric acid(SUA),creatinine(Cr),urea nitrogen(BUN),xanthine oxidase(XOD)levels,and urine uric acid(UUA)level were detected.HE staining was used to observe pathological changes in renal tissue.Immunohistochemical method was used to detect changes in uric acid transporter 1(URAT1)in rat kidney tissue.qRT-PCR was used to detect the expression level of the glucose transporter 9(GLUT9)gene in rat kidney tissue.Results:Compared with the normal control group,the model group rats showed a depressed mental state and increased uri-nation,with higher SUA,Cr,BUN,and XOD,but lower UUA(P<0.05).The epithelial boundaries of renal tubules were blurred in the model group,with cell swelling and degeneration,and inflammatory cell infiltration in the renal interstitium.The expressions of URAT1 protein and GLUT9 mRNA increased in the model group(P<0.05).Compared with the model group,the rats in the allopurinol group and TFCS low,medium,and high-dose groups showed better mental state and urina-tion condition,with lower SUA,Cr,BUN,XOD,but higher UUA(P<0.05).The renal tissue damage was alleviated,and the expressions of URAT1 protein and GLUT9 mRNA were reduced in these groups(P<0.05).Conclusion:TFCS can effec-tively reduce the level of UA in HUA rats and has a protective effect on the kidneys.Its mechanism may be related to inhibi-ting the activity of key enzymes of uric acid synthesis and regulating the expression of uric acid transporter.
