Effects to Euonymus alatus on renal structure and function of diabetic ne-phropathy mice
Aim:To investigate the effects of Euonymus alatus(GJY)on renal structure and function of mice with dia-betic nephropathy.Methods:Ten 7-week-old db/m mice were used as blank group;60 db/db mice were randomly allocated into model group,GJY low,medium,high dose[1.5,3.0,6.0 g/(kg·d)]groups and irbesartan[20 mg/(kg·d)]group,with 12 mice in each group.The blank group and model group received intragastric administration of normal saline,while the other 4 groups received corresponding drugs for 12 weeks.During the experiment,the general state of mice was ob-served,and the fasting blood glucose(FBG),24-hour urine output and water intake were detected before and after adminis-tration.After the last administration,urine albumin/creatinine(UACR),serum creatinine(SCr)and blood urea nitrogen(BUN)were measured;the kidney were weighed to calculate the kidney index(KI),the left kidney tissue samples were stained with HE,PAS and Masson,and the histological findings were observed under optical microscope;the right renal cor-tex was isolated and the ultrastructural changes were observed under transmission electron microscope.Results:Compared with model group,KI,FBG,SCr,BUN and UACR of mice in GJY high dose group and irbesartan group decreased(P<0.05),mesangial matrix percentage and collagen area percentage decreased(P<0.05),renal pathological damage was alle-viated,the thickness of kidney basement membrane decreased(P<0.05),and the fusion of foot process improved.Concl-usion:High dose of GJY can effectively reduce blood sugar levels,repair kidney function and structure,and has good thera-peutic effects on diabetic nephropathy.
