Exploring characteristic genes and immune infiltration features of dengue based on bioinformatics
Aim:To investigate the characteristic genes and immune infiltration features of dengue.Methods:Based on the GEO database gene chips(GSE96656 and GSE206829)and R software,differentially expressed genes(DEG)between patients with dengue and healthy people were analyzed.Characteristic genes were screened by random forest model and LAS-SO regression model.And the gene expressions were validated with external datasets.CIBERSORT algorithm was adopted to perform the immune cell infiltration analysis of dengue,and the correlations between characteristic genes and immune cell infiltration were also analyzed.Results:A total of 500 DEG were obtained,Random forest model and LASSO regression model pointed out IFI44L and FMO4 as characteristic genes.External gene chip validation revealed that IFI44L showed a high expression in dengue patients.IFI44L was positively correlated with the infiltration degree of M1 macrophages,activated dendritic cells and memory B cells(P<0.05).FMO4 was positively correlated with the infiltration degree of M1 macropha-ges and activated memory CD4+T cell(P<0.05).Conclusion:IFI44L and FMO4 have the potential to serve as character-istic genes of dengue and are associated with immune cell infiltration such as M1 macrophages infiltration.
