Karyotype analysis and genome survey of Lindera megaphylla
[Objective]In order to clarify the chromosome morphology and genomic basic information of bigleaf spicebush(Lindera megaphylla),wild bigleaf spicebush was used as the material in this study.[Method]Fluorescence in situ hybridization(FISH)was employed to analyze the chromosomes and genomic survey was conducted to predict the basic information of the bigleaf spicebush genome.[Result]The chromosome number of bigleaf spicebush was 2n=24,with a karyotype formula of 2n= 2X= 24 = 18m(2SAT)+ 6sm,and one pair of satellite chromosomes on the 9th chromosome,classified as type 2B chromosomes.The absolute length variation range of chromosomes was 2.92-5.83,the relative length variation range was 5.33%-11.11%,and the karyotype asymmetry coefficient was 59.49%.The study also found one pair of 5S rDNA hybridization sites and one pair of 18S rDNA hybridization sites located on the 5th and 9th chromosomes,respectively.The two hybridization signals of 18S rDNA were located at the same position and had similar intensity.The genomic survey results showed that the genome size of bigleaf spicebush was 1.38 Gb,the heterozygosity rate was 0.8%,and the repeat sequence proportion was 70%.[Conclusion]Bigleaf spicebush,a diploid with 24 chromosomes,has a relatively symmetric karyotype.It is also a primitive species in the evolution of Lauraceae,and its chromosomes have not undergone recombination,variation or intraspecific hybridization.Bigleaf spicebush has a big genome size with a relatively high heterozygosity rate and a high proportion of repeat sequences.
