摘要

泰乐菌素发酵过程中,发酵培养基成分对生物组分间的转化起着重要作用.文章通过研究发酵培养基碳源、氮源及前体物质添加对泰乐菌素组分含量的影响,探讨泰乐菌素组分稳定性最优培养条件.其结果表明,在发酵开始 4 h向发酵液中加入 0.15%的三甲铵乙内酯,12 h后添加 0.20%的丙二酸钠,以菜籽油为碳源,异亮氨酸、亮氨酸、缬氨酸按1.0∶1.5∶2.0 比例直接作为氮源补料,发酵液pH值为 7.0 时,可明显提升泰乐菌素A组分含量,降低大菌素C含量,使总组分含量达到稳定.

Abstract

In the process of tylosin fermentation,the composition of fermentation medium plays an important role in the transformation of biological components.In this paper,the effects of carbon source,nitrogen source and precursor addition on the content of tylosin components in fermentation medium were studied,and the optimal culture conditions for the stability of tylosin components were discussed.The results showed that 0.15%trimethylammonium glycolide was added to the fermentation broth at the beginning of 4h,and 0.20%sodium malonate was added after 12 h.Taking rapeseed oil as the carbon source,isoleucine,leucine and valine were directly used as nitrogen source supplement at the ratio of 1.0∶1.5∶2.0,and the pH of the fermentation broth was 7.0,the content of tylosin A component was significantly increased,the content of macrocin C was reduced,and the total component was stable.
