摘要

以头孢菌素C生产菌株顶头孢霉菌为研究对象,研究了多种氮源对头孢菌素C产量的影响.通过多因子二水平分析实验、爬坡实验和响应面法优化发酵培养基,确定最佳氮源为蛋氨酸、玉米蛋白粉、豆油,最佳发酵培养基配方:蛋氨酸为 0.93 g/L、玉米蛋白粉为 4.96 g/L、豆油为 6.33 g/L.在此发酵配方下,头孢菌素C产量可达 33.21 g/L,较初始配方提高 16.24%.

Abstract

With the strain of recombinant cephalosporium acremonium producing cephalosporin C as the research object,the effect of multiple nitrogen sources on the production of cephalosporin C was studied.Through multi-factor two-level analysis experiment,climbing experiment and response surface method to optimize the fermentation medium,the optimal nitrogen sources were methionine,corn protein powder,and soybean oil.The optimal fermentation medium formula was:0.93 g/L of methionine,4.96 g/L of corn protein powder,and 6.33 g/L of soybean oil.Under this fermentation formula,the yield of cephalosporin C could reach 33.21 g/L,which was 16.24%higher than the original formula.
