研究达克替尼玉米醇溶蛋白纳米粒(DAC-ZNPs)最佳制备工艺,并对其制剂学性质进行表征.采用相分离法制备DAC-ZNPs,用正交实验法筛选处方工艺,用HPLC法测定达克替尼的含量,激光粒度仪测定纳米粒的粒径,透射电镜观察纳米粒的外观形态,用超滤法测定包封率.按最优处方工艺制备的DAC-ZNPs为球形粒子,平均粒径为(145±25)nm,粒度分布均匀,DSC结果表明药物是以无定形状态分散于纳米粒中,包封率为90.16%士 1.34%,体外24 h累积释放度为52.06%.采用相分离法制备的DAC-ZNPs粒径分布均匀,药物包封率高,具有一定的缓释效果,该制备工艺流程简单、重现性好.
Preparation and characterization of Dacomitinib zein nanoparticles
The optimal preparation technology of Dacomitinib Zein nanoparticles(DAC-ZNPs)was studied and their pharmaceutical properties were characterized.DAC-ZNPs were prepared by phase separation method,and the optimum prescription was screened out by orthogonal design.The content of dacomitinib was determined by HPLC.Granulometer was applied to determine particle size and distribution.Transmission electron microscope was used to observe DAC-ZNPs morphology,and the encapsulation rate was measured by ultrafiltration method.DAC-ZNPs prepared under optimum conditions were spherical grains,with the average particle size of(145±25)nm,which were distributed evenly.DSC results indicated that the drug was dispersed in nano-particles in a non-crystalline state,with an entrapment efficiency of 90.16%±1.34%.The in vitro drug release was 52.06%in 24 hours,which was followed by a sustained release.The particle size distribution of DAC-ZNPs prepared by phase separation method was uniform,and the drug encapsulation efficiency was high.DAC-ZNPs had certain sustained-release effects.The preparation process is simple and reproducible.
