紫外-可见分光光度法测定赣南风车子中总黄酮的含量

摘要
目的:建立赣南风车子中总黄酮的含量测定方法.方法:采用超声提取法提取赣南风车子中的总黄酮,以芦丁为对照品,采用紫外-可见分光光度法测定总黄酮含量,并进行方法学考察.结果:总黄酮含量与吸光度(508 nm)呈现良好的线性关系,线性回归方程为y=8.652x-8.524E-4,R2=0.9986,精密度、重复性、稳定性良好,平均加样回收率97.74%,RSD为1.84%(n=6).结论:建立的以硝酸铝为显色剂的紫外-可见分光光度法测定赣南风车子中总黄酮含量的方法快速、准确,操作简单,专属性强,可作为其含量测定的方法.
Abstract
OBJECTIVE:To establish a method for the determination of total flavonoids in Gannan Combretum alfredii.METHODS:The total flavonoids in Gannan Combretum alfredii were extracted by ultrasonic extraction,and the total flavonoids content was determined by ultraviolet-visible spectrophotometry with rutin as the standard reference,and the methodological investigation was also performed.RESULTS:The total flavonoids content showed a good linear relationship with absorbance(508 nm),the linear regression equation was y=8.652x-8.524E-4,R2=0.9986,with good precision,reproducibility and stability,the average addition recovery and RSD was 97.74%,1.84%(n=6),respectivity.CONCLUSIONS:The established ultraviolet-visible spectrophotometry with aluminium nitrate as chromogenic reagent for the determination of total flavonoids in Gannan Combretum alfredii presented in the experiment is rapid,accurate,simple to operate and specific,and can be used as a method for its determination.
