淡黑雷松藻低聚糖硫酸酯的制备及抗凝血活性研究

摘要
以淡黑雷松藻(Lessonia nigrescens)为原料,采用分步提取和定向改性的方法获得低聚糖硫酸酯ACV1,对其进行了糖醛酸含量、硫酸基含量、相对分子质量、单糖组成理化性质分析,采用傅里叶变换-红外光谱(FT-IR)和核磁共振碳谱(13C-NMR)对其结构进行了表征.体外抗凝血试验结果显示,ACV1可显著延长活化部分凝血活酶时间(APTT)、凝血酶原时间(PT)和凝血酶时间(TT)(P<0.05),减少纤维蛋白原(FIB)的生成(P<0.05).体内斑马鱼试验结果显示,ACV1可显著加快斑马鱼的节间血管血流速度(P<0.01).ACV1作为一种天然活性原料具有良好的抗凝血活性,有潜力改善高凝血状态的药品、日用化妆品等功能性产品的开发.
Abstract
In this study,oligosaccharide sulfate ACV1 was obtained from Lessonia nigrescens by step-by-step extraction and directional modification.physicochemical properties of ACV1 were analyzed,including uronic acid content,sulfate content,relative molecular weight,and monosaccharide composition.the structure of ACV1 was characterized using fourier transform Infrared Spectroscopy(FT-IR)and Nuclear magnetic resonance carbon Spectroscopy(13C-NMR).In vitro anticoagulation experiments showed that ACV1 significantly prolonged activated partial thromboplastin time(APTT),prothrombin time(PT),and thrombin time(TT)(P<0.05),and reduced the production of fibrinogen(FIB)(P<0.05).In vivo,zebrafish experiments revealed that ACV1 significantly accelerated the blood flow velocity in the intersegmental vessels of zebrafish(P<0.01).As a natural active ingredient,ACV1 possesses good anticoagulant activity and has the potential to be applied in the development of functional products such as drugs and daily cosmetics for improving hypercoagulable states.
