A Method for Improving the Separation Degree of Isomers of Moxifloxacin Hydrochloride
Based on the Agilent Poroshell EC-C18 chromatography column(4.6 mm× 100 mm,2.7 µm),the authors studied a method to improve the separation of moxifloxacin hydrochloride and its enantiomers.The results showed that when mobile phase A was an aqueous solution containing ammonium acetate,copper sulfate,and L-isoleucine,with ammonium acetate concentration of 50 mmol·L-1,copper sulfate concentration of 5 mmol·L-1,L-isoleucine concentration of 10 mmol·L-1,mobile phase B was methanol,the volume ratio of mobile phase A to mobile phase B was 70∶30,the flow rate was 1.0 mL·min-1,the injection amount was 10 µL,the column temperature was 40 ℃,and the detection wavelength was 293 nm,the resolution of moxifloxacin hydrochloride isomers reached 6.4.This method did not use chiral chromatography columns,and had a better separation moxifloxacin hydrochloride and its isomers compared to existing technologies.
