一种提高盐酸莫西沙星异构体分离度的方法

基于Agilent Poroshell EC-C18色谱柱(4。6 mm×100 mm,2。7 μm),作者研究了提高盐酸莫西沙星及其对映异构体分离度的方法。结果表明,当流动相A为含有醋酸铵、硫酸铜和L-异亮氨酸的水溶液,且醋酸铵浓度为50 mmol·L-1,硫酸铜的浓度为5 mmol·L-1,L-异亮氨酸的浓度为10 mmol·L-1,流动相B为甲醇,流动相A与流动相B体积之比为70∶30,流速为1。0mL·min-1,进样量为10 µL,柱温为40 ℃,检测波长为293 nm时,盐酸莫西沙星对映异构体分离度达到6。4。该方法不使用手性色谱柱,相比现有技术,可以更好地分离盐酸莫西沙星及其对映异构体。
A Method for Improving the Separation Degree of Isomers of Moxifloxacin Hydrochloride
Based on the Agilent Poroshell EC-C18 chromatography column(4.6 mm× 100 mm,2.7 µm),the authors studied a method to improve the separation of moxifloxacin hydrochloride and its enantiomers.The results showed that when mobile phase A was an aqueous solution containing ammonium acetate,copper sulfate,and L-isoleucine,with ammonium acetate concentration of 50 mmol·L-1,copper sulfate concentration of 5 mmol·L-1,L-isoleucine concentration of 10 mmol·L-1,mobile phase B was methanol,the volume ratio of mobile phase A to mobile phase B was 70∶30,the flow rate was 1.0 mL·min-1,the injection amount was 10 µL,the column temperature was 40 ℃,and the detection wavelength was 293 nm,the resolution of moxifloxacin hydrochloride isomers reached 6.4.This method did not use chiral chromatography columns,and had a better separation moxifloxacin hydrochloride and its isomers compared to existing technologies.
