摘要

为了明确冷应激对民猪肌肉组织抗氧化相关基因表达的影响,试验选取1~2月龄的民猪仔猪9头,随机分为3组,分别饲养在常温(22~25℃,对照)、短期低温(4 ℃ 3 d)和长期低温(4 ℃15 d)环境下.通过实时荧光定量PCR的方法检测常温组、短期低温组和长期低温组民猪仔猪的背最长肌中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPX)基因、核因子E2相关因子2(Nrf2)、Kelch样环氧氯丙烷相关蛋白-1(Keap1)mRNA表达量的变化.结果表明:与常温对照组相比,短期低温组和长期低温组民猪肌肉组织中SOD基因表达量变化不显著(P>0.05),而GPX表达量显著升高(P<0.05);短期低温组Nrf2基因表达量显著升高(P<0.05),但是长期低温组降低;短期低温组Keap1基因表达量变化不显著(P>0.05),而长期低温组中其表达量显著下降(P<0.05).说明冷应激对民猪肌肉组织抗氧化物相关基因的表达有一定的影响.

Abstract

The aim of this experiment was to study the effect of cold stress on the expression of antioxidant-related genes in muscles of Min pigs.Nine Min piglets at 1-2 months were randomly divided into three groups,which were fed at room temperature(22~25 ℃),short-term cold exposure(4 ℃ for 3 d)and long-term cold exposure(4 ℃ for 15 d),respectively.The mRNA expression levels of superoxide dismutase(SOD),glutathione peroxidase(GPX),Nuclear factor erythroid 2-related factor 2(Nrf2)and Kelch-like ECH-associated protein1(Keap1)in longissimus dorsi muscle of piglets in room temperature group,short term cold exposure group and long term cold exposure group were detected by quantitative real-time PCR.The results showed as follows:Compared with the room temperature control group,no significant difference in the expression of the SOD gene.However,the significant increase in GPX expression(P<0.05)was detected in the short-term cold exposure group and the long-term cold exposure group.The expression level of Nrf2 gene was significantly increased in the short-term cold exposure group(P<0.05)and decreased in the long-term cold exposure group.There was no significant difference in Keap1 gene expression between short-term cold exposure group and room temperature group,but it was significantly decreased in long-term cold exposure group(P<0.05).The results indicated that cold stress could impact the expression of antioxidant-related genes in muscles of Min pigs.
