零下低温降温平衡对辽宁绒山羊细管冻精品质的影响

在制作辽宁绒山羊细管冻精时,降温平衡是重要的技术环节.为了提高辽宁绒山羊细管冻精质量,试验将降温平衡的温度范围设定为-1~-2℃低温,并利用这种低温降温平衡方法制作细管冻精(试验组),与利用常规降温平衡(温度0℃)方法制得的细管冻精(对照组)进行了品质(原精活力、冻前活力、冻后活力、冻后 37℃存活时间、精子畸形率)比较;选取 3~5 周岁、1.5~2.5 周岁、10~12 月龄母羊,将每个年龄段母羊分为试验组和对照组,其中试验1,2,3 组的细管冻精采用低温降温平衡方法制得,对照1,2,3 组的细管冻精采用常规降温平衡方法制得,分别比较试验1 组和对照1 组、试验2 组和对照2 组、试验3 组和对照3 组母羊的情期受胎率.结果表明:试验组与对照组的原精活力、冻前活力、精子畸形率间差异不显著(P>0.05),试验组冻后活力和冻后37℃存活时间显著高于对照组(P<0.05);试验组母羊情期受胎率均高于对照组,其中试验1 组显著高于对照1 组(P<0.05).说明在制作辽宁绒山羊细管冻精时可以将降温平衡的温度设定为-1~-2℃,有利于提高细管冻精的质量和情期受胎率.
The effect of low temperature vitrification on the quality of fine tubes of Liaoning cashmere goat semen
Cooling balance is an important technical step in the production of Liaoning cashmere goat frozen semen.In order to improve the quality of Liaoning cashmere goat fine tube frozen semen,the temperature range of cooling balance was set to-1 to-2℃,and this low temperature cooling balance method was used to prepare fine tube frozen essence(experimental group).The quality of fine tube frozen sperm(0℃,control group)was compared with that obtained by conventional cooling balance method,including original sperm vitality,pre-freezing vitality,post-freezing vitality,survival time at 37℃ after freezing,and malformed sperm rate.Ewes aged 3 to 5 years old,1.5 to 2.5 years old and 10 to 12 months old were selected and divided into control groups and experimental groups.The fine tubes frozen semen of experimental groups 1,2 and 3 were obtained by low temperature cooling balance method,and the fine tube frozen semen of control groups 1,2 and 3 was obtained by conventional cooling balance method.The estrus conception rate between experimental group 1 and control group 1,experimental group 2 and control group 2,experimental group 3 and control group 3 were compared respectively.The results showed as follows:There were no significant differences between the experimental group and the control group in prosperm viability,pre-freezing viability and sperm malformation rate(P>0.05).The post-freezing viability and survival time at 37℃ of the experimental group were significantly higher than those of the control group(P<0.05).The impregnation rate of estrus in experimental groups were higher than that in control groups,and that in experimental group 1 was significantly higher than that in control group 1(P<0.05).This shows that the temperature of cooling equilibrium can be set to-1 to-2℃ when making Liaoning cashmere goat fine tube frozen essence,which is conducive to improving the quality of fine tube frozen essence and the transmission and delivery rate.
