Identification and study on the pathogen of common bean anthracnose
In order to confirm the pathogenic species leading to common bean anthracnose in the region of Hulan district,Harbin city,Heilongjiang province, the strain WZL01 was obtained by isolation and culture.Through the comprehensive identification of morphology and molecular biology, the strain was determined to be Colletotrichum spaethianum(C.spaethianum).The biological characteristics of the strain by mycelial growth method and microscopic examination method were studied. The results show that the strain is suitable for growth in 25~30 ℃,pH=6~8,dark conditions, the appropriate medium is potato dextrose ager(PDA)and common bean leaf medium, the appropriate carbon source is maltose, and the nitrogen source is ammonium sulfate. The fungicide virulence test is conducted in the room, and the results show that 8 fungicides such as carbendazim, and the median effect concentration(EC50)values of 40%difenoconazole and 50%carbendazim are 0.031 8 and 0.481 3,respectively,showing better inhibitory effects. The pathogen, biological characteristics and applicable chemical agents of common bean anthracnose in the common bean planting area of Hulan District,Harbin City,Heilongjiang Province are clarified, which lays a research foundation for the future screening of local disease-resistant varieties and field control.
