Quantitative determination of melanin in Auricularia auricula by H2O2 oxidation-fluorescence
A method for the determination of melanin in Auricularia heimuer by H2O2 oxidation fluorescence analysis was established and the content of melanin in Auricularia auricula was determined.The conditions for the determination of fluorescence produced by the oxidation of melanin in Auricularia auricula were screened and the detection limit,precision,stability and standard addition recovery experiments were evaluated.The melanin in Auricularia auricula has an optimal excitation wavelength of 340 nm and an optimal emission wavelength of 460 nm.When the concentration of NaOH is 0.15 mol L-1,the concentration of H2O2 is 10%,the oxidation temperature is 60 ℃,the oxidation time is 1.5 h,and the ultrasonic time is 40 min,the concentration of melanin in the range of 200~1 200 μg·mL-1 shows a good linear correlation with the fluorescence intensity,and R2 of the linear regression equation is 0.996 0,the limit of detection is 0.92 μg·mL-1,and the average recovery is 102.75%.The stability and precision are 1.19%and 1.29%,respectively,and the content of melanin in Auricularia auricula is 2.10%.The above results indicate that the H2O2 oxidation-fluorescence method can quantify the melanin in Auricularia auricula accurately and conveniently.
