Identification and analysis of UPL family genes of E3 ubiquitin ligases responding to salt stress in sugar beet
Sugar beet is a biennial herbaceous plant and an important sugar crop which is widely grown in northeast,northwest and north of China.Using sugar beet T710-Mu line as an experimental material,three UPL family genes containing HECT domain in response to salt stress were obtained by establishing hidden Markov model screening database with the aid of sugar beet transcriptome database under salt stress.The three UPL proteins,BvUPL3,BvUPL4 and BvUPL5,were analyzed for their physical and chemical properties,evolutionary relationships,conserved structural domains and protein interaction networks.BvUPL3,BvUPL4 and BvUPL5 are predicted to be distributed in Chr09,Chr07 and Chr08,respectively,and have typical HECT-type structural domains.The identified UPL genes encoding E3 ubiquitin ligase belong to the UPL family genes.The predicted protein interactions suggest that these UPL proteins interact with each other and may interact with other components of the ubiquitin proteasome system,such as ubiquitin-coupled factors,E2 ubiquitin ligases and the 26S proteasome on proteins.Combining the results of fluorescence quantitative PCR and transcriptome data,the relative expression of BvUPL3 gene is significantly increased in leaves and roots compared with the control group,and the relative expression of BvUPL4 and BvUPL5 genes is significantly down-regulated in leaves and increased in roots compared with the control group.These results can provide a theoretical basis for the subsequent in-depth study of sugar beet ubiquitin ligases.
