一株野生乳孔硫磺菌Laetiporus cremeiporus的分离鉴定及其生物学特性研究

在东北林业大学校园内采摘到一株野生硫磺菌子实体,经分离纯化培养后,利用分子生物学分析,基于内转录间隔区(Internal transcribed spacer,ITS)序列建立系统发育树,鉴定其为乳孔硫磺菌(Laetiporus cremeiporus,L.cremeiporus).采用单因素实验研究了不同碳源、氮源、pH和温度对菌丝生长的影响,乳孔硫磺菌菌丝生长的最适碳源为葡萄糖,最适氮源为硫酸铵,最适pH为4.0,最适温度为20℃.根据单因素实验结果,设计了正交实验,乳孔硫磺菌菌丝最适生长因子组合为葡萄糖、牛肉膏、pH 4.0、30 ℃,各因素对菌丝生长的影响程度为pH>温度>碳源>氮源.
Isolution,identification and biological characteristics of a wild strain of Laetiporus cremeiporus
A wild mushroomfruiting body was collected from the campus of Northeast Forestry University.After the isolation and purification,based on the ITS sequence,a phylogenetic tree was established by molecular biological analysis,and the fungus was identified as Laetiporus cremeiporus(L.cremeiporus).The effects of different carbon sources,nitrogen sources,pH,and temperatures on the growth of mycelia were studied by single factor experiments.The optimal carbon,nitrogen,pH,and temperature for the mycelial growth of L.cremeiporus are glucose,(NH4)2SO4,4.0,and 20 ℃,respectively.Orthogonal experiments were designed and carried out according to the results of single factor experiments.The results show that the optimal factor composition for the mycelial growth of L.cremeiporus is glucose,beef extract,pH 4.0,and 30 ℃.The degree of various influence factors on mycelial growth is pH>temperature>carbon source>nitrogen source.
