Study on Porcine Sperm 3D Trajectory Extraction Mehod Based on Lensless Diffraction Imaging
Sperm motility detection is an important step in the production process of animal husbandry.The existing method of computer-assisted sperm analysis(CASA),which uses a microscope to capture video of sperm movement,is evaluated based on the planar motion trajecars of sperm obtained by the microscope imaging.In fact,the movement of individual sperm is carried out in three dimensions.In this study,a method of 3D trajectory extraction of pig sperm based on lensless diffraction imaging was proposed.The plane location of sperm head was obtained through reconstruction and image processing of the lensless semen imaging captured.An evaluation function was designed for the accuracy of object image distance parameters in the reconstruction process,and the object image distance of the target particle was locked by self-focusing algorithm.Then the three-dimensional motion trajectories of individual sperm are obtained.The results showed that the proposed method could achieve a planar positioning accuracy of 1.4 μm and a height positioning accuracy of±5 μm for individual sperm,and could obtain a relatively high precision three-dimensional motion trajectory,which is expected to provide more scientific data support for sperm quality assessment and reproductive medicine research.
