黑龙江省不同产区五味子的木脂素及有机酸的含量测定及对比分析研究

目的:对黑龙江省14个产区五味子的五味子醇甲、五味子醇乙、五味子酯甲、五味子甲素、五味子乙素、五味子丙素6种木脂素以及苹果酸和柠檬酸2种有机酸的含量进行测定,为五味子的深入开发和质量控制提供一定的参考依据.方法:参照《中国药典》2020年版测定五味子的水分和总灰分;采用HPLC法测定五味子6种木脂素以及2种有机酸的含量;应运聚类分析方法对黑龙江省14个产区的五味子的测定结果进行分析.结果:不同产区五味子的木脂素和有机酸的含量差别较大.14个产区的五味子均不含五味子酯甲,但是五味子醇甲的含量最高,五味子乙素、五味子醇乙、五味子甲素次之,五味子丙素含量最低;虎林市产的五味子的总木脂素百分含量最高,达到了 1.568%,而清河林业局产的五味子的百分含量最低,仅为0.830%;平原区产的五味子的木脂素总百分含量平均值高于山区产的五味子.五味子中苹果酸和柠檬酸的百分含量分别在1.74~5.87%、15.51~21.29%之间,有机酸总含量最高的产区是清河林业局,最低的产区是虎林市,百分含量分别为26.41%、19.04%.山区栽培种五味子的有机酸总百分含量平均值高于平原区产的五味子.当分类聚类为11时,可将14个产区的五味子聚为四大类,可有效区分山区与平原区的五味子.结论:当以木脂素的总含量最为评价指标时,平原产的五味子的质量略优于山区产的五味子.但是以有机酸为评价指标时,山区产的五味子的质量略优于平原产的五味子.在实际应用中,可以根据具体的需求选择对应产区的五味子.
Objective:The contents of schisandrol A,schisandrol B,schisantherin A,schisandrin A,schisandrin B,schisandrin C,malic acid and citric acid in Schisandra chinensis(Turcz.)Baill from 14 producing areas in Heilongjiang Province were determined,so as to provide a certain reference basis for the further development and quality control of Schisandra chinensis(Turcz.)Baill.Methods:The contents of water and total ash of Schisandra chinensis(Turcz.)Baill were determined according to the 2020 edition of Chinese Pharmacopoeia;the HPLC method was used to determine the contents of 6 lignans and 2 organic acids;the results of Schisandra chinensis(Turcz.)Baill from 14 producing areas in Heilongjiang Province were analyzed by cluster analysis.Results:The contents of lignans and organic acids in Schisandra chinensis(Turcz.)Baill from different producing areas were quite different.Schisandra chinensis(Turcz.)Baill from 14 producing areas did not contain schisantherin A,but the content of schisandrol A was the highest,followed by schisandrin B,schisandrol B,schisandrin A,and schisandrin C was the lowest.The total lignan content of Schisandra chinensis(Turcz.)Baill produced in Hulin is the highest,reaching 1.568%,while that of Schisandra chinensis produced by Qinghe Forestry Bureau is the lowest,which is only 0.83%.The average content of total lignans of Schisandra chinensis(Turcz.)Baill produced in plain area was higher than that in mountainous area.The percentage contents of malic acid and citric acid in Schisandra chinensis(Turcz.)Baill ranged from 1.74 to 5.87%and 15.51 to 21.29%,respectively.The highest total organic acid content was Qinghe Forestry Bureau,and the lowest was Hulin City,with a percentage content of 26.41%and 19.04%,respectively.The average content of total organic acid of Schisandra chinensis(Turcz.)Baill cultivated in mountain area is higher than that in plain area.When the classification clustering was 11,Schisandra chinensis(Turcz.)Baill from 14 producing areas can be grouped into four categories,which can effectively distinguish Schisandra chinensis(Turcz.)Baill from mountain area and plain area.Conclusion:When the total content of lignans is the most evaluation index,the quality of Schisandra chinensis(Turcz.)Baill produced in plain is slightly better than that in mountainous area.However,when using organic acid as the evaluation index,the quality of quality of Schisandra chinensis(Turcz.)Baill produced in mountainous area is slightly better than that in plain.In practical application,Schisandra chinensis(Turcz.)Baill can be selected according to the specific needs.
