Correlation Between Learning and Cognitive Ability and Neuropeptide Y,Cortisol in Mice Under Chronic Unpredictable Mild Stress
Objective To investigate the correlation between the learning and cognitive ability of mice under chronic unpredictable mild stress(CUMS)and the levels of serum neuropeptide Y(NPY),cortisol and other stress factors.Methods A total of 12 C57BL/6 mice were divided into control group and CUMS group,with 6 mice in each.The mice in CUMS group completed 30 days of CUMS modeling,while the mice in control group received no special treatment,and the changes in body mass index of the two groups of mice were monitored regularly.Morris water maze was used to detect the movement track,latency,two-quadrant/four-quadrant distance ratio and the times of crossing the platform in two groups of mice.Serum NPY,cortisol α-amylase and tyrosine levels were detected by enzyme-linked immunosorbent assay(ELISA).Results At the 9th day after modeling,the body mass index of mice in CUMS group reduced than that of the control group(P<0.05),and the mice tended to be irritable,which suggested that the stress measures were effective and the model was successfully established.On the 2th day of Morris water maze,there were no significant differences in the movement track,latency,two-quadrant/four-quadrant distance ratio and the times of crossing the platform between two groups of mice(P>0.05);on the 6th day of Morris water maze,there were significant differences in the movement track,latency,two-quadrant/four-quadrant distance ratio and the times of crossing the platform between two groups of mice(all P<0.05).Compared with the control group,the levels of serum NPY and cortisol of mice in CUMS group in-creased,and the differences were statistically significant(all P<0.05).A linear relationship existed between the serum NPY level and the two-quadrant/four-quadrant distance ratio of mice(y=-0.001 x+1.738,R2=0.586,P=0.004),and a linear relationship existed between serum NPY level and latency of mice(y=-0.027x+51.130,R2=0.498,P=0.010).Conclusion Under the CUMS state,the increase of serum NPY level in mice is related to the improvement of learning and cognitive ability.
