Relationship Among the Expressions of miR-204 and miR-504 in Hypopharyngeal Squamous Cell Carcinoma Tissue and Clini-copathological Features and Prognosis
Objective To investigate the relationship among the expressions of microRNA-204(miR-204),miR-504 in hypopharyngeal squamous cell carcinoma(HSCC)tissue and clinicopathological features and prognosis.Methods A total of 122 HSCC patients who were admitted to author's hospital from April 2018 to April 2020 were selected.The intraoperative cancer tissues and cancer adjacent tissues(≥2 cm from the tumor resection margin)were collected,the rel-ative expression levels of miR-204 and miR-504 in cancer tissues and cancer adjacent tissues were detected by reverse tran-scription polymerase chain reaction(RT-PCR).The relationship between the expressions of miR-204,miR-504 and the clinicopathological features of HSCC patients was analyzed.HSCC patients were followed up for 3 years.The prognostic factors of HSCC patients were analyzed by univariate and multivariate Cox proportional hazard regression models.Kaplan-Meier survival curve was drawn,3 years overall survival of HSCC patients with low expression and high expression of miR-204 and miR-504 were analyzed.Results Compared with adjacent tissues,the expression of miR-204 and miR-504 in HSCC cancer tissue significantly decreased(all P<0.05).There was no statistically significant difference in the expres-sions of miR-204 and miR-504 among HSCC patients with different gender,age,smoking history,alcohol consumption history,T stage,site of onset,the number of lymph nodes,extranodal extension and postoperative chemoradiation(P>0.05),the expressions of miR-204 and miR-504 in N1-N3 stage and poorly differentiated HSCC patients were significant-ly lower than that in N0 stage and medium to high differentiated HSCC patients(all P<0.05).The Cox regression analy-sis results showed that elevated T and N stages,positive lymph nodes>2,extranodal extension,low differentiation,decreased miR-204 expression and de-creased miR-504 expression were risk factors for poor prognosis in HSCC patients(P<0.05).During a 3-year follow-up period,5 out of 122 HSCC patients were lost and 117 were ultimately followed up.The Kplan-Meier survival curve analysis showed that the 3-year overall survival rates in the miR-204 and miR-504 low expression groups were lower than those in the high expression group[54.29%(38/70)vs.70.21%(33/47),P<0.05;50.68%(37/73)vs.77.27%(34/44),P<0.05].Conclusion miR-204 and miR-504 are both low expressed in HSCC tissues,and their low expression is associated with elevated T and N stages,poor differentiation and poor prognosis in HSCC patients.
