Study of in vitro Antimicrobial Efficacy and Preparation of Compound Chlorhexidine Acetate Spray
Objective To investigate the preparation of compound chlorhexidine acetate spray and its in vitro anti-microbial activity.Methods The compound chlorhexidine acetate spray was prepared.The paper-disc agar-diffusion method was used to determine the minimum inhibitory concentration(MIC)of each component of compound chlorhexidine acetate spray against 4 standard strains:Pseudomonas aeruginosa,Staphylococcus aureus,Clostridium sporogenes and Candida albicans;the same method was used to detect the bacteriostatic activity of compound chlorhexidine acetate spray against 4 standard strains with chlorhexidine acetate solution(self-made formulation from the NO.980 hospital)and met-ronidazole solution(0.05 mg/ml)as the control.Results A total of 3 batches of the compound chlorhexidine acetate spray were prepared in this study,the chlorhexidine acetate content of labeled amount acetate in the compound chlorhexi-dine acetate spray was 99.81%,101.67%and 103.09%,respectively;the content of polyvinyl alcohol was 102.62%,100.18%,and 104.43%of the labeled content,respectively;the content of metronidazole was 100.89%,103.33%,and 102.82%of the labeled content,respectively,the quality test results of 3 batches of compound chlorhexidine acetate spray all complied with the requirements.The diameter of inhibition zones of compound chlorhexidine acetate spray and chlorhexidine acetate solution against Candida albicans,Staphylococcus aureus and Pseudomonas aeruginosa was signifi-cantly increased than that of metronidazole solution(all P<0.05);the diameter of inhibition zones of compound chlo-rhexidine acetate spray and metronidazole solution against Clostridium sporogenes was significantly increased than that of chlorhexidine acetate solution(all P<0.05).Conclusion The effective antimicrobial components in the formulation of compound chlorhexidine acetate spray were chlorhexidine acetate and metronidazole,with no antagonistic effect observed when mixed with other components in the formulation.Compared with typical single-component disinfection antimicrobial agents,the compound chlorhexidine acetate spray exhibits broad-spectrum antimicrobial advantages.
