Key Genes and Potential Targets Screening of Renal Calculi Formation based on Bioinformatics Methods
Objective To study the key differential genes,signaling pathways and protein-protein interaction(PPI)networks of renal calculi,so as to find potential diagnosis and treatment targets,and provide new diagnosis and treatment ideas for refractory,recurrent and hereditary high-risk stones.Methods Based on bioinformatics methods and R soft-ware,the gene chip data sets GSE73680 and GSE117518 were removed from batch effects and standardized.The key genes and potential targets of renal calculi formation were screened by gene difference analysis,gene ontology(GO)/Kyoto encyclopedia of genes and genomes(KEGG)analysis and weighted gene co-expression network analysis(WGC-NA).Results Differential gene analysis showed that 28 genes were significantly down-regulated and 33 genes were sig-nificantly up-regulated,and there was a co-expression relationship between up-regulated genes and down-regulated genes.Enrichment analysis showed that multiple differential genes acted on the development of kidney,collecting duct,renal tu-bule and ureter,and MMP3 gene played a central role.WGCNA obtained four hub genes,namely BFSP1,ITGB3,OG-DHL,and UCHL1.Conclusion MMP3,BFSP1,ITGB3,OGDHL and UCHL1 genes may be potential targets for the formation of renal calculi Randall plaques,which can provide the target of gene detection and the direction of follow-up treatment for rare and relapsed refractory renal calculi.
