山楂提取物经猴头菌转化后的产物对四氧嘧啶诱导的1型糖尿病小鼠的干预效果

目的 探究猴头菌(Hericium erinaceus)转化山楂提取物(hawthorn fruit extract,HFE)的产物对四氧嘧啶诱导的1型糖尿病(diabetes mellitus type 1,T1DM)小鼠的干预作用及可能机制.方法 60只4周龄雄性KM小鼠按随机数字表法分为转化组、猴头菌发酵液+1%HFE组、猴头菌发酵液组、1%HFE组、阳性组、对照组,每组10只.对照组小鼠正常饲养;其余各组建立四氧嘧啶诱导下的T1DM小鼠模型.造模成功后,转化组小鼠每天用20ml/kg试样1[猴头菌接种于1%HFE(W/V)培养基经发酵离心后取上清液]灌胃;猴头菌发酵液+1%HFE组小鼠每天用20 ml/kg试样2[HFE溶于猴头菌发酵液离心后的上清液,配制浓度为1%(W/V)]灌胃;猴头菌发酵液组小鼠每天用20 ml/kg试样3[猴头菌发酵液离心后取上清液]灌胃;1%HFE组小鼠每天用20 ml/kg试样4[HFE溶于生理盐水,配制浓度为1%(W/V)]灌胃;阳性组小鼠每天用20 ml/kg试样5[盐酸二甲双胍溶于生理盐水,配制浓度为4g/L]灌胃;对照组小鼠每天用20 ml/kg生理盐水灌胃,均连续灌胃10天.利用紫外分光光度法检测各组小鼠血清中随机血糖、餐后血糖和果糖胺水平;采用称重法检测小鼠体质量;采用余量法检测小鼠饮水量.结果 灌胃后,转化组、阳性组小鼠随机血糖、餐后血糖、果糖胺水平均明显低于猴头菌发酵液+1%HFE组、猴头菌发酵液组和1%HFE组,而体质量明显升高(P均<0.05).结论 HFE经猴头菌生物转化后的产物可能通过缓解胰岛素抵抗,改善胰岛β细胞功能,消除血糖累积,增加葡萄糖利用率,降低血浆渗透压,抑制非酶糖基化反应来降低血糖、果糖胺,从而有效干预四氧嘧啶诱导的T1DM.
Effect of Hawthorn Fruit Extract Transformed by Hericium Erinaceus on Alloxan-Induced Diabetes Mellitus Type 1 Mice
Objective To investigate interventional effect and possible mechanism of transformation product about hawthorn fruit extract(HFE)by Hericium erinaceus on alloxan-induced diabetes mellitus type 1(T1DM)mice.Methods A total of 60 4-week-old male KM mice were randomly divided into transformation group,Hericium erinaceus fermentation broth+1%HFE group,Hericium erinaceus fermentation broth group,1%HFE group,positive group and control group,with 10 mice in each.Mice in control group were fed normally;the others groups were established as T1DM mice models by alloxan induction.After successful modelling,mice in transformation group were given 20 ml/kg of specimen 1[supernatant of Hericium erinaceus inoculated in 1%HFE(W/V)medium after fermentation and centrifu-gation]by gavage per day;mice in Hericium erinaceus fermentation broth+1%HFE group were given 20 ml/kg of specimen 2[HFE was dissolved in the supernatant of Hericium erinaceus fermentation broth after centrifugation,the concentration was 1%(W/V)]by gavage per day;mice in Hericium erinaceus fermentation broth group were given 20 ml/kg of specimen 3(supernatant of Hericium erinaceus fermentation broth after centrifugation)by gavage per day;1%HFE group mice were given 20 ml/kg of specimen 4[HFE was dissolved in normal saline,the concentration was 1%(W/V)]by gavage per day;mice in positive group were given 20 ml/kg of specimen 5(metformin hydrochloride was dis-solved in normal saline,the concentration was 4 g/L)by gavage per day;mice in control group were given 20 ml/kg nor-mal saline per day by gavage,and all mice were given continuous gavage for 10 days.The serum levels of random blood glucose,postprandial blood glucose and fructosamine in mice were measured by UV spectrophotometry;the body mass of mice were measured by weighing method;the water intake of mice was measured by residual method.Results After ga-vage,the levels of random blood glucose,postprandial blood glucose,fructosamine of mice in transformation group and positive group were significantly lower than those in Hericium erinaceus fermentation broth+1% HFE group,Hericium erinaceus fermentation broth group and 1%HFE group,while the body mass increased significatly(all P<0.05).Conclusion The products of HFE after the biotransformation of Hericium erinaceus may improve pancreatic β-cell function,remove glucose accumulation,in-crease glucose utilisation,decrease plasma osmolality,inhibit non-enzymatic glycosylation reactions to decrease blood glu-cose and fructosamine levels by alleviating insulin resistance,so as to effectively intervene in alloxan-induced T1DM.
