摘要
目的 为了探讨不同浓度大麻二酚(CBD)对A549非小细胞肺癌的增值抑制和凋亡的相关影响.方法 以不同浓度CBD(0、10、20、40μmol/L)作用细胞24h后,采用CCK-8法检测A549细胞的活力;采用克隆形成实验检测细胞的增值能力;应用流式细胞术检测CBD对A549细胞凋亡和活性氧(ROS)含量的影响.构建A549皮下异种移植瘤模型,探讨不同浓度CBD对小鼠皮下肿瘤的影响,采用苏木精-尹红染色法和原位末端转移酶标记技术检测移植瘤组织细胞凋亡.结果 体外实验表明,不同浓度CBD均对A549细胞的增值有抑制作用,并且存在剂量依赖性;流式细胞术结果显示,随着CBD药物浓度升高,细胞凋亡数目增多以及ROS含量增高.进一步体内实验结果显示,与对照组相比,实验组皮下移植瘤组织中细胞凋亡增多且对小鼠重要脏器没有药物毒性.结论 CBD在体内外均能诱导A549细胞凋亡、抑制其增值,具有潜在抗肺癌细胞的作用.
Abstract
Objective To investigate the effects of different concentrations of cannabidiol(CBD)on cell prolifera-tion and apoptosis of A549 non-small cell in lung cancer.Metheds After acting on the cells with different concen-trations of CBD(0,10,20,40μmol/L)for24h,the viability of A549 cells was detected by the CCK-8 assay,and the proliferation of the cells was detected by the clone formation assay.Flow cytometry was applied to detect the effects of CBD on apoptosis and reactive oxygen species(ROS)content in A549 cells.The A549 xenograft tumor model was constructed to explore the effects of different concentrations of CBD on subcutaneous tumors in mice,and the apoptosis of transplanted tumor tissues was detected by hematoxylin-eosin staining and in situ terminal transferase labeling methods.Results In vitro experiments showed that different concentrations of CBD exhibited antiproliferative action to A549 cells in a dose-dependent manner.Flow cytometry results showed that the higher CBD drug concentrations was,the more increase in the number of apoptotic cells as well as in ROS content.Fur-ther vivo experiments showed that the apoptosis in the subcutaneously grafted tumor tissues of the experimental group increased and there was no drug toxicity to the vital organs of the mice,compared with the control group.Conclusion CBD exhibited antiproliferative and apoptotic effects on A549 cells in both vivo and ex vivo,and therefore,had a potential anti-lung cancer cell effects
