宽叶荨麻中总黄酮的提取工艺及活性研究

以宽叶荨麻为原料,乙醇为提取剂,采用超声波提取法,对宽叶荨麻中总黄酮的提取工艺条件及其体外活性进行了研究.采用单因素实验考察了超声时间、乙醇浓度、料液比以及超声次数对宽叶荨麻总黄酮提取量的影响,同时采用正交试验对提取工艺进行优化.结果表明:宽叶荨麻中总黄酮最佳提取工艺为70%乙醇在料液比1∶50,超声20 min,超声2 次,此条件下平均提取率为12.21%.0.6 g/L总黄酮对DPPH·自由基和ABTS+自由基清除活性较强,1.5 g/L总黄酮对α-淀粉酶和α-葡萄糖糖苷酶也有一定的抑制作用,具有一定的开发价值.
Study on Extraction Process and Activity of Total Flavonoids in Urtica Latifolia
Using urtica latifolia as raw material and ethanol as extractant,the extraction conditions and in vitro activity of total flavonoids from urtica latifolia are studied by ultrasonic extraction.The effects of ul-trasonic time,ethanol concentration,solid-liquid ratio and ultrasonic times on the extraction of total fla-vonoids from urtica latifolia are investigated by single factor experiment,and the extraction process is op-timized by orthogonal experiment.The results show that the optimum extraction process of total flavonoids in Urtica latifolia is 70%ethanol at a solid-liquid ratio of 1∶50,ultrasonic 20 min,ultrasonic 2 times,the average extraction rate is 12.21%.The total flavonoids of 0.6 g/L has strong scavenging activity on DPPH·free radical and ABTS+free radical,and 1.5 g/L total flavonoids also has certain inhibitory effect on α-amylase and α-glucosidase,which has certain development value.
