Effects of Glutamine Dipeptide on Proliferation and Apoptosis of Bovine Small Intestinal Epithelial Cells
[Objective]This study is conducted to investigate the effects of glutamine dipeptide on the proliferation and apoptosis of bovine small intestinal epithelial cells.[Method]Bovine small intestinal ep-ithelial cells were cultured in vitro and subjected to Glutamine(Gln)starvation treatment.Gln,Alanyl-glutamine(Ala-Gln),or Glycine-glutamine(Gly-Gln)were added respectively.Cell viability,rate and cycle of apoptosis,overflow amount of lactic dehydrogenase(LDH),and expression levels of apoptotic-related proteins(Bax,Bcl-2,and Caspase-3)were measured.[Result]The results showed as follows:compared with CK(the Gln starvation group),the bovine small intestinal epithelial cells supplemented with Ala-Gln and Gly-Gln significantly increased cell viability and significantly decreased LDH amount(P<0.05).In addition,compared with CK,the addition of Gln,Ala-Gln and Gly-Gln significantly de-creased the rate of G0/G1 and G2/M phase retarded cells and increased the rate of S phase cells(P<0.05);The apoptosis rate was significantly decreased(P<0.05);The expression of anti-apoptotic factor Bcl-2 was significantly increased,while the expressions of pro-apoptotic factor Bax and Caspase-3 were significantly decreased(P<0.05).[Conclusion]In conclusion,glutamine dipeptides(Ala-Gln and Gly-Gln)can promote the proliferation and reduce the apoptosis of bovine small intestinal epithelial cells,thus protecting the integrity of the intestinal mucosal structure,which can provide a theoretical basis for the application of glutamine dipeptide in bovine intestinal nutrition.
