Study on the protective effect of Pueraria Seabuckthorn granules on acute alcohol-induced liver injury
Objective To study the protective effects of Pueraria Seabuckthorn granules on acute alcohol-induced liver injury model in mice.Methods Sixty kunming mice were randomly divided into blank control group,model control group and dose groups(including three groups,low,medium and high dose groups,and the doses of Pueraria Seabuckthom granules for the three groups were 0.25,0.50 and 1.50 g/kg,respectively.).Each group had 12 mice.Except from the blank control group,acute alcoholic liver injury model was established in other groups.The mice in the dose groups were given oral gavage with corresponding doses of Pueraria Seabuckthorn granules for 30 days.The contents of malondialdehyde(MDA),glutathione(GSH)and triglycerides(TG)in liver issue of the mice at the end of the experiment were measured,and the pathologic examination of hepatic tissue was conducted.Results The contents of MDA,GSH and TG in liver and pathological scores in the model group were significantly different from those in the blank control group(all P<0.05),indicating that the model of acute alcoholic liver injury was successfully constructed.In the high dose group of Pueraria Seabuckthom granules,MDA,GSH and TG contents in liver homogenate of mice were(2.44±0.39)nmol/mg,(0.25±0.07)mg/g and(0.022±0.007)mmol/g,respectively,and pathological score was(2.75±0.97).In the model control group,MDA,GSH and TG contents were(3.27±0.68)nmol/mg,(0.17±0.09)mg/g and(0.029±0.005)mmol/g,respectively,and pathological score was(3.83±0.39).Compared with the model control group,the contents of MDA and TG in liver tissue of alcohol-induced liver injury mice were decreased and the contents of GSH were increased,and hepatocyte steatosis score decreased in the high-dose group of Pueraria Seabuckthorn granules(all P<0.05).Conclusions Pueraria Seabuckthorn granules has the protective effects on acute alcohol-induced liver injury in mice.
