Monitoring results of SARS-CoV-2 in urban sewage in Zhengzhou city
Objective To analyze the monitoring results of SARS-CoV-2 in environmental sewage in Zhengzhou city,Henan province,provide data support for environmental monitoring,and preliminarily explore the role of environmental monitoring in COVID-19 prevention and control.Methods Two sewage treatment plants in Zhengzhou city were selected as monitoring sites.From March-July 2023 each sewage treatment plant collected two samples of sewage per week;the enrichment was conducted by PEG method.Multiple real-time fluorescence reverse transcription-polymerase chain reaction(RT-PCR)method was used to detect and analyze novel coronavirus;the whole genome sequence was obtained by SARS-CoV-2 sequencing,and the virus type was judged by analysis software.Results The total detection rate of novel coronavirus in urban sewage was 89.09%.From March to April,the detection rate of novel coronavirus was 76.47%and 81.82%respectively,and from May to July,the detection rate was 100.00%.Of the 55 sewage samples collected,SARS-CoV-2 was detected in sewage solution,sediment,supernatant and concentrated samples.Virus content size:concentrated sample>stock solution>supernatant>sediment.N gene calculation showed that the concentration of SARS-CoV-2 in concentrated samples was 13.57 times that in sewage solution.The concentration of SARS-CoV-2 in urban sewage peaked in the 23rd week,then began to decline,and then dropped to a low point in the 26th week.In May,the top two viruses in sewage were XBB.1.9,XBB.1.16 and its sub-branches.Conclusions The detection rate of SARS-CoV-2 in environmental sewage in Zhengzhou is high,especially from May to July.The virus type is Omicron XBB subbranch,and environmental monitoring can be used as a supplement to hospital case monitoring.
