Study on Evaluation of Acute Toxicity and Antitumor Activity of Different Chinese Medicinal Monomers
Objective:To investigate the acute toxicity of five Chinese medicinal monomers:curcumin,baicalein,betulinic acid,resvera-trol,and dihydroartemisinin,and to preliminarily explore their antitumor activities.Methods:The IncuCyte live-cell imaging technology was used to assess tumor cell proliferation,and flow cytometry was employed to detect tumor cell apoptosis.The acute toxicity of different Chinese medicinal monomers was preliminarily evaluated through a mouse acute toxicity test.Healthy 8-week-old female B-NDG mice were used to establish patient-derived xenograft(PDX)models of gastric cancer,colorectal cancer,and breast cancer.The antitumor ac-tivities of curcumin,baicalein,resveratrol,and dihydroartemisinin were compared using intraperitoneal and intravenous injection meth-ods.Results:All four Chinese medicinal monomers:curcumin,baicalein,resveratrol,and dihydroartemisinin,showed varying degrees of inhibition of tumor cell proliferation and promotion of tumor cell apoptosis(P<0.05),with curcumin exhibiting the strongest activity.Acute toxicity tests indicated that mice had the highest tolerance to baicalein,followed by curcumin.Intravenous injection of curcumin demonstrated significant antitumor activity in the BP0880 gastric cancer PDX model(P<0.05).Conclusion:Among the five mono-mers,curcumin exhibits the strongest antitumor activity and shows a relatively high tolerance in acute toxicity tests.
