目的 探究结直肠癌(CRC)癌组织中miR-106a、miR-92a、miR-31与其新生血管生成及侵袭转移的关系.方法 选取本院2018年1月至2020年1月诊治的100例结直肠癌患者作为CRC组,另选取同期健康体检者60例作为对照组.全部均行实时荧光定量聚合酶链反应(RT-PCR)检测miR-106a、miR-92a、miR-31表达量,采用免疫组化法检测CD34表达,计算微血管密度(MVD);术后随访3年,收集患者肿瘤侵袭转移等资料.分析miR-106a、miR-92a、miR-31与CRC新生血管生成及侵袭转移的关系.结果 CRC组miR-106a、miR-92a、miR-31表达量及MVD均比对照组高(P<0.05).Pearson相关性分析显示,miR-106a、miR-92a、miR-31表达量与CRC患者MVD均呈正相关(P<0.05).单因素分析显示,侵袭转移组肿瘤高、中分化程度人数及TNM分期Ⅲ、Ⅳ期人数比未侵袭转移组多,术前miR-106a、miR-92a、miR-31表达量比未侵袭转移组高(P<0.05).多元逐步回归分析显示,肿瘤分化程度、TNM分期、术前miR-106a表达量、术前miR-92a表达量、术前miR-31表达量均为CRC患者肿瘤侵袭转移的独立影响因素(P<0.05).结论 结直肠癌癌组织中miR-106a、miR-92a、miR-31高表达,且与结直肠癌新生血管生成及侵袭转移密切相关.
Objective To investigate the relationship between miR-106a,miR-92a,miR-31 and neovascularization,invasion and metastasis in colorectal cancer(CRC).Methods 100 patients with colorectal cancer diagnosed and treated in our hospital from January 2018 to January 2020 were selected as CRC group,and another 60 healthy patients during the same period were selected as control group.Real-time fluorescence quantitative polymerase chain reaction(RT-PCR)was used to detect the expression levels of miR-106a,miR-92a and miR-31,and immunohistochemical method was used to detect the expression of CD34,and microvascular density(MVD)was calculated.The data of tumor invasion and metastasis were collected after 3 years of follow-up.The relationship between miR-106a,miR-92a and miR-31 and CRC neovascularization,invasion and metastasis was analyzed.Results The expression levels and MVD of miR-106a,miR-92a and miR-31 in CRC group were higher than those in control group(P<0.05).Pearson correlation analysis showed that the expression levels of miR-106a,miR-92a and miR-31 were positively correlated with MVD in CRC patients(P<0.05).Univariate analysis showed that the number of patients with high and moderate differentiation and the number of patients with TNM stage Ⅲ and Ⅳ in the invasive metastasis group was higher than that in the non-invasive metastasis group,and the preoperative expression levels of miR-106a,miR-92a and miR-31 were higher than those in the non-invasive metastasis group(P<0.05).Multiple stepwise regression analysis showed that the degree of tumor differentiation,TNM stage,preoperative miR-106a expression level,preoperative miR-92a expression level,and preoperative miR-31 expression level were all independent influencing factors of tumor invasion and metastasis in CRC patients(P<0.05).Conclusion miR-106a,miR-92a and miR-31 are highly expressed in colorectal cancer tissues,and are closely related to angiogenesis,invasion and metastasis of colorectal cancer.
