A Study on Genetic Polymorphism of Qingzhou Silver Melon and its Related Species Using RAPD
Qingzhou silver melon has excellent quality,sweetness,juiciness,and strong aroma,making it one of the national famous and high-quality varieties of melons and vegetables.Using RAPD marker technology,genomic analysis was conducted on 9 test materials of Qingzhou silver melon and its closely related species,and clustering analysis and principal coordinate analysis graphs of the 9 test materials were constructed.The amplification results showed that among the selected 30 RAPD random primers,24 primers amplified a total of 276 bands,of which 176 bands were polymorphic,with an average of 7.3 polymorphic bands obtained per primer and an average polymorphism rate of 63.77%.The results of clustering analysis and principal coordinate analysis indicated that the genetic similarity coefficients of the 9 test materials ranged from 0.61 to 0.98,suggesting certain genetic differences among the test materials.Among them,M1(three-leaf crispy flower skin melon)had a smaller genetic similarity coefficient with other test materials,indicating a more distant relationship;while M5(Qingzhou silver melon),M4(white-skinned crispy sweet horn honey),and M6(fire phoenix silver melon)had higher genetic similarity coefficients,indicating that these three germplasm are the most closely related.
