本研究旨在解决花生粉中黄曲霉毒素B1能力验证样品均匀性差的问题.通过对花生粉采用正己烷除脂后进行干燥处理,再加入混有待测物的乙醇溶液,经过混匀、干燥、粉碎、过筛后进行分装.深入研究了除脂比例、风干温度与时间对检测结果的影响.结果显示,当花生粉与正己烷质量比为1:2,在60 ℃下风干12 h的条件下,样品的检测结果呈现出高度的均一性,且稳定性最佳.在此条件下制备了两批浓度分别为10μg/kg和20 μg/kg的样品,通过三重四极杆液相色谱质谱联用仪进行分析,并采用同位素内标法进行评估.样品的添加回收率在85%以上,具有良好的均匀性和稳定性,完全符合能力验证样品的要求.
Development and Analysis of Aflatoxin Bi Proficiency Testing Samples of Peanut Powder
This study aims to solve the problem of poor uniformity of aflatoxin B1 proficiency tes-ting samples of peanut powder.The peanut powder was dried after being degreased with n-hexane,then the ethanol solution mixed with the substance for test were added.After mixing,drying,crush-ing,and sieving,the peanut powder was repackaged.The effects of degreasing ratio,air-drying tem-perature and time on the test results were thoroughly investigated.The results showed that when the mass ratio of peanut powder to n-hexane was 1:2 and air-dried at 60 ℃ for 12 hours,the test results of the samples exhibited a high degree of uniformity and the best stability.Under these conditions,two batches of samples with concentrations of 10 μg/kg and 20 μg/kg respectively were prepared.They were analyzed by triple quadrupole liquid chromatography-mass spectrometry and evaluated by the isotope internal standard method.The standard recovery rate of the samples was above 85%,with good uniformity and stability,the samples were fully met the requirements of proficiency testing.
