A Rapid Detection Method Forpigment Based on Ps-Co-P4VP/AU Peroxidase Activity
Polystyrene-poly(4-vinylpyridine)core-shell microspheres were synthesized by lotion polymeri-zation with styrene and 4-vinylpyridine as monomer. It was combined with gold nanoparticles to form poly-styrene-poly(4-vinylpyridine)/gold(PS-co-P4VP/Au)composite nanosphere with spherical morphology, favorable dispersity. In the presence of hydrogen peroxide,3,3',5,5'-tetramethylbenzidine(TMB)can be catalyzed by the microspheres with peroxidase activity to generate blue products. The optimum pH of the material for adsorbing basic orange 21 was 9.It was found that the peroxidase activity of microspheres will be decreased after adsorption, and the decrease is linear with the pigment concentration, based on the phe-nomenon,a rapid detection method for basic orange 21 was established. When the pH=9,TMB concentra-tion was 1.4 mmol/L,H202 concentration was 140 mmol/L,and material concentration was 2.5 mg/ml,the detection effect is the best. And under this conditions, the detection limit of basic orange 21 was 0.6 μg/mL,the recovery rate in the wine and sugar was 92.2%~102.8%.
