茯苓对大鼠肾源性水肿的影响

为研究茯苓对大鼠肾源性水肿的影响,探讨茯苓治疗肾源性水肿的可能机制,将150只SD级大鼠随机分为对照组(12只)和模型组(138只),使用阿霉素对模型组大鼠建立肾源性水肿模型.随机将肾源性水肿模型组大鼠分为非给药模型组,茯苓粉(生、熟、代培)低、高剂量组,茯苓醇提物低、中、高剂量组,茯苓水提物低、中、高剂量组.每组8只,雌雄各半,连续21 d灌胃给药.第0,7,14,21天记录体重变化,第21天取血,通过Elisa实验检测大鼠血清IL-2、IL-6、ANP及BNP水平.结果发现,茯苓粉(生、熟、代培)对于水肿程度的缓解(体重)和尿蛋白的降低表现出一定作用.茯苓醇提物对于肾源性水肿大鼠血清ANP、IL-6的降低效果最为显著,5种茯苓样品都可以降低肾源性水肿大鼠血清BNP、IL-2的水平.茯苓可以缓解大鼠肾源性水肿,并降低大鼠血清ANP、BNP、IL-2、IL-6的水平,其中高剂量的熟茯苓对肾源性水肿大鼠尿蛋白缓解效果最好,茯苓醇提物对于肾源性水肿大鼠血清ANP、IL-6的降低效果最为显著,说明茯苓对肾源性水肿的治疗具有一定的积极作用.
Preliminary Study on the Effect of Poria Cocos on Nephrogenic Oedema in Rats
This paper aims to study the effect of Poria cocos on nephrogenic edema in rats,and to explore the possible mechanism of its treatment.150 SD rats were randomly divided into control group(12)and model group(138),and the model was established using doxorubicin.Rats with renal oedema were ran-domly divided into non-administered model group,with low and high doses of Poria cocos powder(raw,cooked,and cultured),low,medium and high alcohol extract,and 8 in each group,half of each male and female,for 21d continuously.Weight changes were recorded on day 0,7,14,21,and blood was harvested on day 21,and serum IL-2,IL-6,ANP,and BNP levels were measured by the Elisa assay.Results show that Poria cocos powder(raw,cooked,and subcultured)had a certain effect on the reduction of oedema degree(weight)and urinary protein,among which cooked Poria cocos powder has the best effect.Poria cocos alcohol extract had the most significant effect on reducing serum ANP and IL-6 in rats with nephro-genic oedema,and all five Poria cocos samples could reduce the levels of serum BNP and IL-2 in rats with nephrogenic oedema.Poria cocos can relieve nephrogenic edema in rats and reduce the levels of serum ANP,BNP,and IL-2 and IL-6.Among them,high dose of cooked Poria cocos had the best effect on relie-ving urinary protein in rats with nephrogenic edema,and Poria alcohol extract had the most obvious effect on reducing serum ANP and IL-6 in rats with nephrogenic edema.indicating that it has a positive effect in the treatment of nephrogenic edema.
