Discussion on the Mechanism of Compound Astragalus Tongluo Decoction in the Treatment of Diabetic Peripheral Neuritis
This study aims to investigate the key active components and pharmacological mechanisms of the Compound Astragalus Tongluo decoction in treating diabetic peripheral neuritis.The ingredients of the Compound Astragalus Tongluo decoction were analyzed by the ultra-performance liquid chromatography-high-resolution mass spectrometry(UPLC-MS/MS),its main signaling pathways for the treatment of dia-betic peripheral neuritis were predicted by network pharmacology.In experimental verification,60 KM mice were selected for DPN modeling,the general situation,blood glucose value,and heat sensitivity val-ue were recorded,mouse sciatic nerve tissues were HE stained,and protein level expression of PI3K,P-AKT and AKT were detected.There were 141 compounds identified by UPLC-MS/MS analysis,which were divided into 11 categories,and most of them were phenolic acids;Network pharmacology research showed that 70 compounds were active components,and the biological processes involving were mainly metabolic regulated by PI3K/AKT signaling pathway.Animal verification experiments showed that there was no significant effect on the expression level of PI3K in the mouse sciatic nerve tissue;however,the phosphorylation level of AKT was significantly decreased in the administered group(p<0.05).The Com-pound Astragalus Tongluo decoction for the treatment of diabeticperipheral neuritis via multi-compo-nents,multi-targets,and multi-pathways,and laid a foundation for the deeply development and applica-tion of this prescription.
