摘要

目的:分析肝细胞癌(HCC)组织中microRNA(miRNA)的表达,构建预后模型,探索肝癌新型诊断及预后相关的miRNA生物标志物.方法:采用R包"edgeR"从TCGA数据库和GEO数据库中获取数据.通过COSMIC分析HCC-Altered Genes的相互作用,使用LASSO Cox回归方法,我们得到4个与预后相关的miRNA,通过构建预后模型预测肝癌预后.结果:在本研究中,7个miRNAs被确定为肝细胞癌的潜在诊断作用.此外,通过筛选TCGA数据库分期相关的miRNA,5个miRNAs与总生存期(OS)相关,3个miRNAs与无进展生存期(PFS)相关.最后将诊断相关和分期相关的miRNAs进行OS相关性分析,筛选出4个miRNAs构建预后模型,对HCC的预后进行预测.这些miRNAs和潜在的通路将成为HCC治疗的靶点.结论:本研究发现4个候选miRNAs可作为HCC生物的标志物和潜在的治疗靶点.

Abstract

Objective:To explore the novel biomarkers of miRNAs of HCC diagnosis and prognostic by analyzing the expression of microRNA(miRNA)in hepatocellular carcinoma(HCC)tissues and es-tablish a prognostic model.Methods:R package"edgeR"was used to obtain data from the TCGA and the GEO database,and COSMIC was used to analyze the interactions of HCC-Altered Genes.Using the LASSO Cox regression method,we obtained 4 miRNAs to the prognosis.Results:In this study,7 miRNAs were identified as a potential diagnostic role for hepatocellular carcinoma.5 miRNAs were associated with overall survival(OS)and 3 miRNAs were associated with progression-free survival(PFS)by screening the staging related miRNAs in the TCGA database.Moreover,this prognosis model,including 4-miRNA signature risk score,was identified as an independent risk factor.These candidate miRNAs and pathways might serve as potential therapeutic targets for HCC treatment.Conclusion:In this study,a total of 4 miRNAs are identified as novel prognostic indicators and thera-peutic targets in HCC patients.
