Expression of ferroptosis-related markers in the anterior vaginal wall of female patients with stress urinary incontinence
Objective:To explore the expression of ferroptosis-related indicators in the anterior vaginal wall tissues of stress urinary incontinence(SUI)patients.Methods:A total of 24 patients undergoing SUI surgery in Renmin Hospital of Wuhan University were selected as the SUI group,and 21 patients who underwent hysterectomy for benign tumors served as the control group.The Fe2+ content and the ex-pression of glutathione peroxidase 4(GPX4)in the vaginal anterior wall were detected by inductively coupled plasma-optical emission spectrometer(ICP-OES)and immunohistochemistry.The levels of malondialdehyde(MDA),reduced glutathione(GSH)and oxidized glutathione disulfide(GSSG)in the two groups were detected by MDA kit,GSH kit and GSSG kit,respectively.And Prussian blue stain-ing was used to detect tissue iron content.The ultrastructure of tissue and cells was observed by trans-mission electron microscope.Results:The content of Fe2+ in the anterior vaginal wall of the SUI group was significantly higher than that of the control group(P<0.000 1),and the expression of GPX4 was significantly lower than that of the control group(P<0.000 1).The levels of MDA and GSSG were significantly higher than those of the control group(P<0.000 1),while the levels of GSH were lower than those of the control group(P<0.000 1).Typical changes of ferroptosis were detected by transmission electron microscopy in the mitochondria of anterior vaginal wall cells in the SUI group.Conclusion:Ferroptosis is involved in the occurrence of SUI and may be an important regulato-ry factor in the process of SUI.
