目的:探究三七叶总皂苷(TSPNL)对癫痫(EP)大鼠miRNA-204和海马神经元损伤的影响.方法:将75只SD大鼠随机分为空白对照组、EP组、TSPNL组、TSPNL+Antagomir-NC组、TSPNL+Antagomir-miR-204组,每组 15只.采用锂-毛果芸香碱制备EP大鼠模型.给予相应的干预后,观察大鼠行为学变化;使用脑电图(EEG)监测大鼠脑电图改变情况;RT-qPCR检测海马组织miR-204、原肌球蛋白相关激酶B(TrkB)、白细胞介素(IL)1β、IL-6的mRNA表达;HE染色检测海马组织病理学变化;免疫荧光法(IF)检测海马组织神经元特异核蛋白(NeuN)阳性细胞数;流式细胞仪检测海马神经元细胞凋亡率;Western Blot检测海马组织凋亡相关蛋白B细胞淋巴瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)、半胱天冬氨酸蛋白酶3(Caspase-3)表达.结果:与EP组比,TSPNL组和TSPNL+Antagomir-NC组大鼠的Racine评分、90 min内癫痫发作次数和持续时间、棘慢波数目、海马组织TrkB mRNA、IL-1β mRNA和IL-6 mRNA表达、神经元细胞凋亡率、Bax和Caspase-3表达降低(P<0.05),海马组织miR-204表达、海马CA1、CA3区NeuN阳性细胞数、Bcl-2表达升高(P<0.05),海马CA1区、CA3区神经元损伤明显减轻;且miR-204的拮抗剂可明显加重海马神经元损伤,逆转TSPNL的抗EP作用.结论:TSPNL可能通过上调miR-204的表达,保护EP大鼠海马神经元损伤.
Protective effect of Panax notoginseng saponins on hippocampal neurons of epileptic rats by regulating miR-204
Objective:To explore the effects of total saponin of Panax notoginseng leaves(TSPNL)on miRNA-204 and hippocampal neuron damage in epileptic(EP)rats.Methods:A total of 75 SD rats were randomly divided into the blank control group,EP group,TSPNL group,TSPNL+ Antagomir-NC group,and TSPNL+Antagomir-miR-204 group,with 15 rats in each group.EP rat model was established by lithium-pilocarpine.After the corresponding interventions were given,the behavioral changes of rats were observed;Electroencephalogram(EEG)was used to monitor the changes of EEG;RT-qPCR was used to detect the expression of miR-204,tropomyosin-related ki-nase B(TrkB),interleukin-1β(IL-1β),and interleukin-6(IL-6)mRNA in the hippocampus;HE stain-ing was used to detect the pathological changes of the hippocampus;immunofluorescence(IF)was used to detect the number of neuron-specific nuclear protein(NeuN)positive cells in hippocampus;flow cytometry was used to detect the apoptosis rate of hippocampal neurons;Western blot was used to detect the expression of Bcl-2,Bax and Caspase-3 in the hippocampus.Results:Compared with those in the EP group,the Racine score,the number and duration of seizures in 90 minutes,the num-ber of spike and slow waves,the expression of TrkB mRNA,IL-1β mRNA,and IL-6 mRNA in the hippocampus,the apoptosis rate of neurons,the expression of Bax and Caspase-3 in TSPNL group and TSPN +Antagomir NC group were lower(P<0.05),the expression of miR-204 in hippocampal tissues,the number of NeuN positive cells and the expression of Bcl-2 in hippocampal CA1 and CA3 were higher(P<0.05),the damage of hippocampal CA1 and CA3 neurons was significantly alleviat-ed;the antagonist of miR-204 can significantly aggravate the damage of hippocampal neurons and re-verse the anti-EP effect of TSPNL.Conclusion:TSPNL may protect hippocampal neurons from EP injury by up-regulating the expression of miR-204.
