An improved method for the establishment of the high concentration of 4NQO induced mouse tongue cancer model
Objective:To investigate an efficient and reliable method for establishing a mouse model of tongue cancer within a short time while maintaining a high survival rate,through a new approach of combined drinking water which contains a high concentration of 4-nitroquinoline-1-oxide(4NQO)and 4NQO-medicated feed.Methods:Twelve BALB/c mice were randomly divided into a control group(H2O,n=4)and two experimental groups,which included a medium dose group(250 μg/mL 4NQO,n=4)and a high dose group(300 μg/mL 4NQO,n=4).The body weight and tongue mucosa status were recorded.The mice were fed with 4NQO water and/or 4NQO-medicated feed for 10 weeks and were executed at the end of 12 weeks,and tongue mucosa samples were taken for HE staining and pathological observation.Results:Successful modeling of tongue cancer was observed in the mice of the medium-dose group,with three of them showing papillary neoplasia in the tongue mucosa.HE staining revealed the presence of nests and keratinized beads.One mouse from the same group exhibited only leukoplakia-like changes in the tongue mucosa,and the epithelium of tongue mucosa dis-played significantly heterogeneous.All mice in the high-dose group died during the experiment.Con-clusion:The administration of 250 μg/mL 4NQO,combined with medicated feed,enabled the induc-tion of a tongue cancer model in mice within 12 weeks while maintaining a high survival rate.
