Objective:To study the antioxidant effect of resveratrol(RSV)after mechanical stress-related in-jury in vitro,and explore its impact on promoting cell repair of mechanical stress injury through mito-chondrial-related pathways.Methods:Mechanical stress(MS)was performed on mouse fibroblast L929 by four-point bending cell mechanical loading apparatus before the treatment of RSV,and the cell viability,oxidative stress,apoptosis,and mitochondrial function were detected.The treatment of RSV was inhibited by a potent and selective Sirt1 inhibitor,Selisistat(Synonyms:EX-527),to evalu-ate the effect of RSV on mechanical stress injury in vitro.These processes aim to evaluate and ana-lyze the therapeutic effect of RSV on oxidative stress injury induced by mechanical force,and the re-sult showed that it might be associated with improving mitochondrial function.Results:RSV can ef-fectively improve fibroblasts'cell viability,apoptosis level,and oxidative stress after mechanical stress injury.It may also improve the function of cells after MS by improving mitochondrial function,and may exert therapeutic effects through mitochondrial dynamics.After being inhibited by EX-527,the above therapeutic effects can also be weakened.Conclusion:RSV is an effective antioxidant drug for the treatment of mechanical injury of mouse fibroblasts,which is beneficial for drug development and application in the treatment of stress urinary incontinence.
