Piezo1促进卵巢癌的上皮间质转化过程

目的:旨在分析Piezo1表达与肿瘤上皮间质转化(EMT)相关蛋白在卵巢癌患者组织中的表达情况和相关性以及研究Piezo1对卵巢癌细胞(SKOV3、A2780)EMT过程的影响.方法:通过生物信息学GSEA法分析Piezo1激活的卵巢癌相关信号通路.采用免疫组化、Western Blot检测正常卵巢组织和卵巢癌组织中Piezo1蛋白及EMT通路相关蛋白E-cadherin、N-cadherin的表达情况,并分析Piezo1表达量与E-cadherin、N-cadherin表达的相关性、用Yoda1 10 μmol/L处理卵巢癌细胞(SKOV3、A2780)2 d,根据转染慢病毒将卵巢癌细胞(SKOV3、A2780)分为sh-NC(转染sh-NC慢病毒)组、sh-Piezo1组(转染sh-Piezo1慢病毒),然后用Western Blot检测各组SKOV3、A2780细胞中E-cadherin、N-cadherin的表达情况.结果:Piezo1的蛋白表达水平在卵巢癌组织中明显高于正常卵巢组织(P<0.05),且通过生物信息学 GSEA 富集分析发现 Piezo1 在卵巢癌中显著激活 EMT 信号通路(P<0.01),Western Blot及免疫组化结果表明E-cadherin的蛋白水平在卵巢癌组织中显著降低(P<0.05),而N-cadherin的表达水平显著升高(P<0.05).相关性分析表明在卵巢癌组织中Piezo1蛋白表达水平与E-cadherin蛋白表达水平显著呈负相关(r=-0.731 7,P=0.001 5),而与N-cadherin呈正相关(r=0.600 7,P=0.003).Yoda1处理后的SKOV3、A2780细胞中,E-cadherin的表达水平降低(均P<0.05),而N-cadherin的表达水平显著升高(P<0.05,P<0.01).敲低SKOV3、A2780 细胞中的Piezo1 后,E-cadherin的表达水平显著升高(P<0.01,P<0.001),而N-cadherin的表达水平降低(P<0.05,P<0.01).结论:Piezo1的高表达可能与患者的不良预后相关,且可能通过促进上皮间质转化过程参与卵巢癌的发生发展.
Piezo1 boosts the epithelial-mesenchymal transformation process in ovarian cancer
Objective:To analyze the expression and correlation of Piezo1 expression and the related pro-teins of tumor epithelial-mesenchymal transformation(EMT)in the tissues of ovarian cancer patients and to investigate the influence of Piezo1 on the EMT process of ovarian cancer cells(SKOV3 and A2780).Methods:The ovarian cancer-related signaling pathways activated by Piezo1 were analyzed by the bioinformatics GSEA method.Immunohistochemistry and Western Blot were used to detect the expressions of Piezo1 protein and the EMT pathway-related proteins E-cadherin and N-cadherin in normal ovarian and ovarian cancer tissues.The correlation between Piezo1 expression and the expres-sion of E-cadherin and N-cadherin was analyzed.Ovarian cancer cells(SKOV3 and A2780)were treated with Yoda1 10 μmol/L for two days.According to the transfection of lentivirus,the ovarian cancer cells(SKOV3 and A2780)were divided into sh-NC(transfection of sh-NC lentivirus)and sh-Piezo1(Transfection of sh-Piezo1 lentivirus)groups.Then Western Blot was used to detect the ex-pression of E-cadherin and N-cadherin in SKOV3 and A2780 cells of each group.Results:The pro-tein expression level of Piezo1 was significantly higher in ovarian cancer than in normal ovarian tissue(P<0.05),and it was found that Piezo1 significantly activated the EMT signaling pathway in ovarian cancer by bioinformatics GSEA enrichment analysis(P<0.01).Western Blot and immunohistochemi-cal results showed that the protein level of E-cadherin in ovarian cancer tissues was significantly de-creased(P<0.05),while the expression level of N-cadherin was significantly increased(P<0.05).Correlation analysis showed that Piezo1 expression in ovarian cancer was significantly negatively corre-lated with that of E-cadherin(r=-0.731 7;P=0.001 5),and positively correlated with that of N-cadherin(r=0.600 7;P=0.003).In SKOV3 and A2780 cells treated with Yoda1,the expression level of E-cadherin was decreased(both P<0.05),while the expression level of N-cadherin was sig-nificantly increased(P<0.05 and P<0.01,respectively).After Piezo1 knockdown in SKOV3 and A2780 cells,the expression level of E-cadherin was significantly increased(P<0.01 and P<0.001,respectively),and that of N-cadherin was decreased(P<0.05 and P<0.01,respectively).Conclu-sion:The high Piezo1 expression may be related to poor prognosis of patients,and may also play a role in the development of ovarian cancer by promoting the process of EMT.
