摘要
建立了测定益母草中盐酸水苏碱含量的HPLC-RID方法,并测定分析了 14 家药厂的益母草中盐酸水苏碱的含量.结果表明:以Mercury益母草专用柱(250 mm×4.6 mm,5 μm)为色谱柱,以 70%乙腈溶液为流动相、流速 1.0 mL/min、柱温 45℃的条件下采用示差检测器进行检测,盐酸水苏碱在 0.100~0.523 mg/mL浓度范围内呈良好的线性关系;回归方程为y=91519 x+1342.2(R2=0.991 6),平均回收率 100.07%,RSD为 0.71%,且该方法具有重复性好、操作简便、快速、稳定、易于普及等优点;各厂家的益母草药材中盐酸水苏碱含量差异较大,14 家厂家中仅有 1367 和 8656 这 2 家的益母草药材符合药典标准;盐酸水苏碱含量与益母草药材性状具有关联性,市民、药企和中药商等在购买益母草药材时,建议选择性状为叶多、青绿、茎细的益母草药材.
Abstract
A HPLC-RID method was established to determine the content of stachydrine hydrochloride in Leonuri Herba,and then the content of stachydrine hydrochloride in the Leonuri Herba samples from 14 manufacturers was determined.The results indicated that the content of stachydrine hydrochloride demonstrated a good linear relationship within the range of 0.100-0.523 mg/mL using the differential detector with the Mercury column(250 mm×4.6 mm,5 μm),mobile phase of 70:30 acetonitrile-water solution,flow rate of 1.0 mL/min,and column temperature of 45℃.The regression equation was y=91 519 x+1 342.2(R2=0.991 6),and the average recovery rate was 100.07%(RSD of 0.71%).The established HPLC-RID method was characterized by good repeatability,simple operation,rapid determination with stable results,and easy popularization.The content of stachydrine hydrochloride varied significantly among samples from different manufacturers,and only the samples from manufacturers 1367 and 8656 met the standards in the Chinese Pharmacopoeia.The content of stachydrine hydrochloride was associated with the medicinal characteristics of Leonuri Herba.According to the results,we suggested that users,pharmaceutical enterprises,and Chinese medicine manufacturers chose the Leonuri Herba materials with rich green leaves and fine stems.
