Optimization of Fermentation Conditions of Bacillus velezensis XY40-1 by Response Surface Methodology
To reduce the fermentation cost and increase the viable count of Bacillus velezensis XY40-1 in fermentation,this study employed full factorial design,orthogonal design,single factor test,and response surface methodology to optimize the fermentation medium and conditions of this strain.The optimal fermentation medium for XY40-1 was composed of 0.7% glucose,1.0% soybean meal,and 0.5% magnesium sulfate heptahydrate.The fermentation of XY40-1 with an inoculation amount of 1.0% at 36.4 ℃ and 196.4 r/min,and initial pH 7.3 showed the viable count reaching 3.57×109 CFU/mL,which increased by 92.97% compared with that without optimization.The viable count in a 200-L fermenter reached 8.23×109 CFU/mL under the optimized conditions.
