Isolation and Identification of an Endophyte with Killing Effect on Pine Wood Nematodes from Pinus massoniana
To obtain the strains capable of killing pine wood nematodes,we collected the healthy plants,the plants infected with pine wood nematodes but still alive,and the plants infected with pine wood nematodes and dead of Pinus massoniana from Liuyang City,Hunan Province.A total of 27 strains of endophytic bacteria were isolated and subjected to assessment of their nematocidal effects.Furthermore,the efficient antagonistic strains were identified.The results showed that strain H-4 isolated from the infected but still alive plants exhibited a high nematocidal activity.The treatment with the fermentation broth of strain H-4 for 48 h achieved the highest killing effect on pine wood nematodes,with the corrected mortality and clearance rates of pine wood nematodes reaching 97.1% and 89.2%,respectively.Strain H-4 was identified as Bacillus siamensis according to the morphological,physiological,and biochemical characteristics and the phylogenetic tree built based on the 16S rRNA sequence.
