Optimization of extraction process and evaluation of antioxidant activity of total flavonoids from Dandelion
Total flavonoids were extracted from dandelion by ethanol extraction.The effects of ethanol concentration,extraction temperature,extraction time and other factors on the extraction efficiency of total flavonoids were investigated.The extraction process was optimized by orthogonal experiment based on single factor experiment.The optimal conditions were ethanol concentration of 40%,extraction temperature of 70℃,extraction time of 80min,under these conditions,total flavonoids content of dandelion extract was 0.9170mg·mL-1.The antioxidant activity of total flavonoids from dandelion was evaluated by DPPH scavenging experiment and hydroxyl free radical scavenging experiment.The IC50 value of DPPH free radical scavenging was 0.8492mg·mL-1,and the IC50 value of hydroxyl free radical scavenging ability was 1.0717mg·mL-1.The results showed that the flavonoid extract of dandelion had good antioxidant ability in scavenging free radicals.
