PB-BBD响应面法优化头花蓼提取工艺及抗氧化活性研究

目的 利用Plackett-Burman和Box-Behnken响应面法优化头花蓼的总黄酮和总多酚超声提取工艺并研究其抗氧化活性.方法 以头花蓼总黄酮和总多酚的综合得率为指标,通过单因素实验确定各影响因素的范围,再利用Plackett-Burman实验筛选影响综合得率的显著因素,最后采用Box-Behnken响应面法进行工艺优化;并通过考察提取物对DPPH自由基、羟基自由基的清除率和对Fe3+的还原能力来评价头花蓼的抗氧化性活性.结果 影响综合得率的显著因素为超声功率、超声温度和超声时间,最优提取工艺为超声功率160W、超声温度58℃、超声时间38min、乙醇浓度40%、液料比25∶1(mL∶g),头花蓼中总黄酮和总多酚的综合得率为68.79mg·g-1,与模型预测值的相对误差为4.15%;当浓度为5mg·mL-1时,此工艺条件下头花蓼提取物对DPPH自由基和羟基自由基的清除率分别为81.66%、84.93%,总还原能力达到2.48.结论 采用基于PB实验设计和BBD响应面法优化得到的头花蓼总黄酮和总多酚提取工艺简便、稳定、可行,且得到的头花蓼提取物具有较强的抗氧化活性.
Optimizing the extraction of Polygonum capitatum by PB-BBD response surface method and its antioxidant research
BJECTIVE To optimize the ultrasonic extraction process of total flavonoids and total polyphenols from Polygonum capitatum by PB design and BBD response surface metohd,and research its antioxidant effects.METHODS Using the yield of total flavonoids and total polyphenols as the indexes,several influencing factors were investigated and Plackett-Burmandesign was used to screen the significant factors.Box-Behnken response surface method was used to optimize the extraction of Polygonum capitatum.Theantioxidant activity of Polygonum capitatum was evaluated by examining the scavenging rate of DPPH and hydroxyl radicals and its ability to reduce Fe3+.RESULTS The significant factors were ultrasonic power,temperature andtime.The optimal extraction method was as follows:the ultrasonic power of 160W,temperature of 58℃,time of 38min,ethanol concentrations of 40%and liquid-to-material ratio of 25∶1(mL·g-1).Under the optimal condition,the comprehensive yield of total flavonoids and total polyphenols was 68.79mg·g-1,the relative error was 4.15%.The in vitro scavenging rates of the extracts at 5mg·mL-1 against DPPH free radicals and hydroxyl radicals were 81.66%and 84.93%,respectively.The total reducing capacity reached 2.48.CONCLUSION The ultrasonic extraction process of total flavonoids and total polyphenols from Polygonum capitatum optimized by PB-BBD is simple,stable and practicable.The extract of Polygonum capitatum in thi sresearch had a strong antioxidant activity.
