Study on optimization of ultrasound assisted enzymatic extraction of polyphenols from Zizyphus mauritiana Lam.and its antioxidant activity
DBJECTIVE Optimize the ultrasound assisted enzymatic extraction process conditions of Zizyphus mauritiana Lam.polyphenols(ZMP)from Zizyphus mauritiana Lam.and determine the antioxidant activity of ZMP.METHOD Based on the single factor experimental investigation,response surface methodology was designed to optimize the extraction conditions of ZMP using liquid-solid ratio,ethanol volume fraction,ultrasound temperature,ultrasound time,ultrasound power,and enzyme dosage as factors.Study the ability of ZMP to scavenge DPPH radicals(DPPH·)and hydroxyl radicals(·OH).RESULTS The optimal extraction conditions of ZMP ultrasonic assisted enzymatic method were as follows:liquid-solid ratio of 40∶1(mL∶g),ultrasonic temperature of 41℃,ultrasonic power of 304W,enzyme dosage of 4.1%(m/m),ethanol volume fraction of 60%,ultrasonic time of 50min,under these conditions,the average extraction amount of ZMP was(18.41±0.16)mg·g-1,which was close to the predicted value(18.33mg·g-1).The results of antioxidant test showed that the mass concentration of ZMP was 3.0μg·mL-1,the clearance rates of DPPH·,·OH were 75%and 72%,respectively,and both of them were higher than that of VC.CONCLUSION The ultrasonic assisted enzymatic extraction process of ZMP optimized by response surface methodology is stable and feasible,and ZMP has strong antioxidant activity,which can be developed and utilized as an antioxidant.
