HPLC同时测定断节参中告达亭和开德苷元的含量

Simultaneous determination of kidjolanin and caudatin in Cynanchum allichii Wight by HPLC

目的建立断节参中开德苷元、告达亭两个C21 甾体类成份的分析方法.方法采用waters SunFire C18(4.6mm×250mm,5μm)色谱柱,乙腈-0.1%磷酸(43∶57)为流动相;流速:1.0mL·min-1;检测波长:224nm;柱温为 30℃.结果开德苷元、告达亭分别在 0.0109~0.544mg·mL-1、0.0079~0.3955mg·mL-1 范围内与峰面积线性关系良好(r≥0.999 6);平均加样回收率分别为 99.82%、99.80%,RSD分别为 1.01%、1.04%(n=6);精密度、稳定性、重复性实验结果RSD均小于 1.0%.结论所建立的方法简单、准确、稳定,可用于评价断节参药材的质量.
OBJECTIVE To establish an HPLC method for simultaneous determination of two C21 steroids including kidjolanin and caudatin in Cynanchum allichii Wight.METHODS The analysis of this drug was performed on a 30℃thermostatic waters Sunfire C18 column(4.6mm×250mm,5μm),with the mobile phase comprising of acetonitrile-0.1%phosphoric acid(43:57)flowing at 1.0mL·min-1,and the detection wavelength was set at 224 nm.RESULTS Kidjolanin and caudatin had a good linear relationship with peak area in the ranges of 0.0109 to 0.544mg·mL-1 and 0.0079 to 0.3955mg·mL-1,respectively(r≥0.999 6).The average recoveries were 99.82%and 99.80%,and RSD were 1.01%and 1.04%(n=6),respectively.RSD of precision,stability and repeatability test results were all less than 1.00%.CONCLUSION The established method is simple,accurate and stable,and can be used to evaluate the quality of the Cynanchum allichii Wight.
