摘要

目的 优化硒化人参多糖的制备工艺.方法 采用水提纯沉法提取多糖,用硝酸-亚硒酸钠法对多糖进行硒化,在单因素实验的基础上对人参多糖的硒化工艺进行响应面分析,采用紫外和红外光谱进行表征.结果 当温度为 71℃、时间为 6.52h、Na2SeO3 质量为 524mg时,硒化人参多糖中的硒含量可达 4.217mg·g-1,红外谱图上可观察到硒的特征峰.结论 所得工艺条件稳定可靠,可为人参硒化多糖的工艺条件提供理论依据与实验参考.

Abstract

OBJECTIVE To optimize the preparation process of selenium-treated ginseng polysaccharide.METHODS Aqueous purification and precipitation method was used to extract polysaccharide,and sodium selenite-nitrate method was used to selenium-treat polysaccharide.Response surface methodology was used to analyze the selenium-treatment of ginseng polysaccharide on the basis of single factor.The content of selenium was determined by ultraviolet spectrophotometry,and the binding of selenium with polysaccharide was verified by infrared spectroscopy.RESULTS When the temperature was 71℃,the time was 6.52h,and the mass of sodium selenite was 524mg,the selenium content of selenium-treated ginseng polysaccharide could reach 4.217mg·g-1,and the characteristic peak of selenium could be observed in infrared.CONCLUSION The obtained process conditions were stable and reliable,which could provid theoretical basis and experimental reference for the process conditions of selenium-treated ginseng polysaccharide.
