低共熔溶剂优化山里红总皂苷类成分的提取工艺研究

摘要
本文采用超声辅助天然低共熔溶剂提取山里红总皂苷成分,基于单因素实验,通过响应面方法对工艺参数进行优化.结果表明,氯化胆碱和乙醇通过氢键相互作用形成的天然低共熔溶剂提取率优于其他组合,山里红总皂苷成分的最佳提取工艺为:低共熔溶剂(DESs)中氯化胆碱与乙醇的摩尔比为1∶3,药材与DESs的料液比为 1∶8(g∶mL),含水量为 20%,超声时间为 25min.在此工艺条件下,山里红总皂苷组分的提取率为(7.21±0.15)%.与传统溶剂提取方法相比,本方法绿色高效,操作简单,可为天然低共熔溶剂提取中药皂苷类成分提供参考依据.
Abstract
In this paper,ultrasound-assisted extraction of total saponins from Hawthorn by natural eutectic solvent,based on the single factor experiment,the process parameters were optimized by response surface method.The results showed that the extraction rate of natural eutectic solvent formed by hydrogen bond interaction between choline chloride and ethanol is better than that of other combinations.The optimum extraction process of total saponins from Hawthorn is as follows:The molar ratio of choline chloride to ethanol in eutectic solvent(Deep Eutectic Solvents,DESs)was 1∶3,the ratio of material to liquid of medicinal material to DESs was 1∶8,the water content was 20%,and the ultrasonic time was 25min.Under these conditions,the total saponins extracted from Hawthorn was(7.21±0.15)%.Compared with the traditional solvent extraction method,this method is green,efficient,and easy to operate.It can provide a reference basis for extracting saponins from traditional Chinese medicine with natural eutectic solvent.
